Acemetacin is commercially available in the form of capsules and sustained-release capsules (Tilur, Tilur retard). It has been approved in many countries since 1985.

Structure and properties

Acemetacin (C21H18ClNO6, Mr = 415.8 g/mol) exists as a yellow to greenish yellow crystalline powder that is practically insoluble in water. It is a glycolic acid ester prodrug of indomethacin and belongs to the arylacetic acid derivatives.

Effects

Acemetacin (ATC M01AB11) has analgesic, antipyretic, and anti-inflammatory properties. The effects are due to inhibition of the synthesis of prostaglandins by inhibition of cyclooxygenase.

Indications

For the treatment of painful and inflammatory conditions (e.g., osteoarthritis, arthritis, gout).

Dosage

According to the professional information. The regular capsules are taken up to three times daily and the sustained-release capsules are taken up to twice daily with a meal.

Adverse effects

Common to very common adverse effects include nausea, vomiting, headache, drowsiness, dizziness, drowsiness, depression, fatigue, tinnitus, and gastrointestinal symptoms such as upper abdominal discomfort, abdominal pain, loss of appetite, diarrhea, and constipation. Numerous other less common adverse effects are possible.
