Amifostine

Amifostine was commercially available as an infusion preparation (Ethyol). It had been approved in many countries since 1996.

Structure and properties

Amifostine (C₅H₁₅N₂O₃PS, M_r = 214.2 g/mol) is a prodrug that is metabolized in tissue to the free thiol.

Effects

Amifostine (ATC V03AF05) is cytoprotective and a free radical scavenger. It protects against chemotherapy-induced acute and cumulative hematologic toxicity and nephrotoxicity.

Indications

• Chemotherapy: reduction of infection risk and nephrotoxicity.
• Radiotherapy: reduction of dry mouth.
