Cefpodoxime

Products

Cefpodoxime is commercially available as film-coated tablets and as granules for preparation of a suspension (Podomexef, generics). It has been approved in many countries since 1991.

Structure and properties

Cefpodoxime (C15H17N5O6S2, Mr = 427.5 g/mol) is present in drugs as cefpodoxime proxetil. It is an ester prodrug and is rapidly converted to cefpodoxime in the body.

Effects

Cefpodoxime (ATC J01DD13) has bactericidal properties against Gram-positive and Gram-negative pathogens. The effects are based on inhibition of cell wall synthesis.

Indications

For the treatment of bacterial infectious diseases with susceptible pathogens. Cefpodoxime is used to treat respiratory and urinary tract infections.

Dosage

According to the SmPC. The drug usually taken twice daily, 12 hours apart and during a meal.

Contraindications

  • Hypersensitivity, including to cephalosporins.
  • Invasive skin and soft tissue infections
  • Cellulitis in infancy
  • Infants under 2 months

Interactions

Drug-drug interactions are possible with probenecid and diuretics. Drugs that increase gastric pH may decrease bioavailability (antacids, H2 antihistamines, proton pump inhibitors).

Adverse effects

Possible adverse effects include hypersensitivity reactions, blood count changes, headache, weakness, dizziness, tinnitus, sensory disturbances, diarrhea, nausea, vomiting, abdominal pain, pruritus, and rash.
