Effects
Cisapride (ATC A03FA02) has prokinetic properties and improves motility of the digestive tract. The effects are due to agonism at serotonin 5-HT4 receptors and release of acetylcholine in the myenteric plexus. It improves lower esophageal sphincter tone and decreases reflux.
Indications
Cisapride has been used to treat gastroparesis and other conditions.
Interactions
Cisapride is metabolized by CYP3A4 and is therefore susceptible to drug-drug interactions.
Adverse effects
Cisapride can prolong the QT interval and cause life-threatening cardiac arrhythmias. Therefore, it has been withdrawn from the market in many countries.
