Clotiapine

Clotiapine is commercially available in tablet form (Entumin). It has been approved in many countries since 1967.

Structure and properties

Clotiapine (C18H18ClN3S, Mr = 343.87 g/mol) is a dibenzothiazepine. The structurally closely related neuroleptic quetiapine (Seroquel) also belongs to this group of drugs.

Effects

Clotiapine (ATC N05AH06) has adrenolytic, antidopaminergic, anticholinergic, antihistamine, sedative, psychomotor depressant, sleep regulator, anxiety and tension reliever, antipsychotic, and contact enhancer properties.

Indications

  • Schizophrenias
  • Manic-depressive psychoses, mania, manic phases.
  • Anxiety, panic states, inner restlessness, agitation.
  • Withdrawal syndrome in alcohol and drug addiction.
  • Neurotic depersonalization syndrome
  • Agitation, aggressiveness, hyperactivity.
  • Sleep disturbances

Dosage

According to the professional information. The tablets are taken up to three times a day.

Contraindications

  • Hypersensitivity
  • Comatose states, severely decreased brain function.

In those at risk for seizures, shock treatments and abrupt dose changes with clotiapine are contraindicated. Full precautions can be found in the drug label.

Interactions

Drug-drug interactions are possible with centrally depressant drugs and with agents that prolong QT interavll.

Adverse effects

Possible adverse effects include agitation, confusion, anticholinergic effects, extrapyramidal disorders, dullness, blurred vision, orthostatic hypotension, dry mouth, and constipation.
