Doxazosin

Products

Doxazosin is commercially available in the form of sustained-release film-coated tablets (Cardura CR, generic). It has been approved in many countries since 1999.

Structure and properties

Doxazosin (C23H25N5O5, Mr = 451.5 g/mol) is present in drugs as doxazosin mesilate, a quinazoline derivative and a white crystalline powder that is sparingly soluble in water.

Effects

Doxazosin (ATC C02CA04) has vasodilator and antihypertensive properties. The effects are due to antagonism at alpha1-adrenoreceptors.

Indications

For the treatment of hypertension (essential hypertension). Doxazosin is not approved for the treatment of benign prostatic hypertension in many countries.

Dosage

According to the SmPC. Tablets are taken once daily, regardless of meals.

Contraindications

  • Hypersensitivity
  • Occlusive diseases of the digestive tract or esophagus

Interactions

Drug-drug interactions are possible with other antihypertensives and phosphodiesterase-5 inhibitors.

Adverse effects

The most common possible adverse effects include abdominal pain, nausea, dry mouth, bronchitis, cough, headache, dizziness, low blood pressure, palpable heartbeats, rapid pulse, itching, pain, weakness, edema, cystitis, and urinary incontinence.
