Fluoxetine is commercially available as tablets, dispersible tablets, and capsules (Fluctine, generics, USA: Prozac). It has been approved in many countries since 1991.

Structure and properties

Fluoxetine (C17H18F3NO, Mr = 309.3 g/mol) is present in the drugs as fluoxetine hydrochloride, a white crystalline powder that is sparingly soluble in water. It is a racemate and was developed starting from diphenhydramine, which itself also inhibits serotonin reuptake.

Effects

Fluoxetine (ATC N06AB03) has antidepressant properties. The effects are due to selective inhibition of serotonin reuptake in presynaptic neurons. The drug has a long half-life of 4 to 6 days. The active desmethyl metabolite norfluoxetine even has a half-life of up to 16 days.

Indications

For the treatment of depression and bulimia nervosa.

Dosage

According to the prescribing information. The drug is usually taken once a day, or twice a day at higher doses, regardless of meals.

  • Hypersensitivity
  • Combination with MAO inhibitors

Interactions

Fluoxetine has a high potential for interaction. It is a substrate and inhibitor of CYP2D6.
