Flupentixol

Products

Flupentixol is commercially available as film-coated tablets and as a solution for injection (Fluanxol). A fixed combination with melitracene is also available (Deanxit). The drug has been approved in many countries since 1967. See also under melitracene and flupentixol.

Structure and properties

Flupentixol (C23H25F3N2OS, Mr = 434.5 g/mol) is a thioxanthene derivative and a racemate. The salt contained in the tablets, flupentixol dihydrochloride, is present as a white crystalline powder that is very soluble in water. In the solution for injection, flupentixol is esterified with decanoic acid.

Effects

Flupentixol (ATC N05AF01) has antipsychotic, disinhibitory, antianxiety, mood-elevating, and, especially at higher doses, depressant properties. The effects are attributed to antagonism at dopamine and serotonin receptors and alpha1-adrenoreceptors. The mean half-life is approximately 34 hours.

Indications

For the treatment of schizophrenia and psychosis.

Dosage

According to the professional information. The dose is adjusted on an individual basis. Tablets are taken once daily in the morning for maintenance therapy. The injectable solution is injected intramuscularly.

Contraindications

  • Hypersensitivity, including to other thioxanthenes.
  • Central depression
  • Comatose states

Interactions

Drug-drug interactions are possible with centrally depressant drugs, with alcohol, and with agents that prolong the QT interval.

Adverse effects

The most common possible adverse effects include drowsiness, agitation, movement disorders, parkinsonism, and dry mouth.
