Metolazone is commercially available in tablet form as a generic product (Metolazone Galepharm). The original Zaroxolyn is no longer available.
Structure and properties
Metolazone (C16H16ClN3O3S, Mr = 365.8 g/mol) is a quinazoline sulfonamide that is sparingly soluble in water. It is structurally related to the thiazides.
Effects
Metolazone (ATC C03BA08) has diuretic and thiazide-like properties. It inhibits reabsorption of sodium chloride at the distal renal tubule.
Indications
For treatment of edema in renal disease and heart failure.
Dosage
According to the SmPC. The usual daily dose is 2.5 to 5 mg, usually as a single dose.

- Hypersensitivity
- Anuria
- Coma hepaticum
- Precomatose states
- Severe disturbances of the electrolyte balance

Adverse effects
The most common possible adverse effects include nausea, vomiting, constipation, diarrhea, headache, dizziness, and muscle cramps. Other possible side effects include hypersensitivity reactions and hypokalemia.
