Naftifine

Products

Naftifine is commercially available as a gel and cream for external use. The drug has not yet been registered in many countries.

Structure and properties

Naftifine (C21H21N, Mr = 287.4 g/mol) is a lipophilic naphthalene derivative and belongs to the group of allylamines, which includes terbinafine. In pharmaceuticals, it is present as naftifine hydrochloride.

Effects

Naftifine (ATC D01AE22) has antifungal properties against dermatophytes, molds, and yeasts. The effects are based on inhibition of squalene-2,3-epoxidase and thus inhibition of the synthesis of ergosterol, an essential component of the fungal cell membrane. Naftifine also has simultaneous anti-inflammatory and antibacterial effects against Gram-positive and Gram-negative bacteria. Unlike azole antifungals, naftifine does not inhibit lanosterol demethylase.

Indications

For the treatment of fungal skin infections and nail fungus.

Dosage

According to the SmPC. For the treatment of fungal skin infections, the drug is applied locally once a day; for the treatment of fungal nail infections, it is applied twice a day.

Contraindications

Naftifine is contraindicated in case of hypersensitivity. For complete precautions, see the drug label.

Interactions

Drug-drug interactions have not been reported.

Adverse effects

Possible adverse effects include local skin reactions such as a burning sensation, dry skin, redness, itching, irritation, and rarely hypersensitivity reactions.
