Nefopam used to be available in many countries in tablet form (Acupan). It is now no longer commercially available.
Structure and properties
Nefopam (C17H19NO, Mr = 253.3 g/mol) exists as nefopam hydrochloride, a white crystalline powder that is soluble in water. It is a benzoxazocine derivative. Nefopam is not structurally related to other analgesics.
Effects
Nefopam (ATC N02BG06) has analgesic properties. It is active centrally at the level of the brain and spinal cord. The effects are likely due to interaction with neurotransmitter systems. Nefopam inhibits the reuptake of several neurotransmitters. It is also sympathomimetic and anticholinergic. Nefopam has a half-life of approximately 6 hours.
Indications
For symptomatic treatment of pain.
Dosage
According to the professional information leaflet. The tablets are usually taken three times daily.
Contraindications
- Hypersensitivity
- Epilepsy
- Combination with MAO inhibitors

Interactions
Drug-drug interactions have been described with MAO inhibitors, anticholinergics, and tricyclic antidepressants.
Adverse effects
Possible adverse effects include:
- Nausea
- Nervousness, paresthesias
- Dry mouth
- Dizziness, low blood pressure, palpitations.
- Urinary retention
- Indigestion
- Convulsions, tremors, confusion, hallucinations.
