Nepafenac

Products

Nepafenac is commercially available as an eye drop suspension in two different concentrations (Nevanac). It has been approved in many countries since 2008.

Structure and properties

Nepafenac (C15H14N2O2, Mr = 254.3 g/mol) exists as a yellow powder. It is an amide analog and a prodrug of amfenac. It passes rapidly through the cornea and is converted by hydrolases to the active drug amfenac.

Effects

Nepafenac (ATC S01BC10) has anti-inflammatory and analgesic properties. The effects are due to inhibition of prostaglandin synthesis.

Indications

  • For prevention and treatment of postoperative pain and inflammation associated with cataract surgery.
  • To reduce the risk of postoperative macular edema associated with cataract surgery in diabetic patients.

Dosage

According to the drug label. The drops are placed in the eye either once or three times a day, depending on the concentration of active ingredient. The vial must be shaken before use because it is a suspension.

Contraindications

  • Hypersensitivity, including to other NSAIDs

Adverse effects
