Nomegestrol acetate is commercially available as a fixed combination with the natural estrogen estradiol in the form of film-coated tablets (Zoely). It has been approved in many countries since 2012 and entered the market in 2013.
Structure and properties
Nomegestrol acetate (C23H30O4, Mr = 370.5 g/mol) is derived from progestin 19-norprogesterone and progesterone, respectively.
Effects
Nomegestrol acetate (ATC G03AA14) has contraceptive properties. The effects are primarily due to inhibition of ovulation. The drug binds to the progesterone receptor and has antigonadotropic, antiestrogenic, and moderate antiandrogenic properties. Nomegestrol acetate has a long half-life of up to 46 hours.
Indications
For oral hormonal contraception in women 18 years of age and older.
Dosage
According to the professional information. The tablets are taken once a day at the same time of day, regardless of meals.

Interactions
Nomegestrol acetate is metabolized primarily by CYP3A. Appropriate drug-drug interactions are possible, for example with CYP inducers.
Adverse effects
The most common possible adverse effects include acne, weight gain, headache, and abnormal withdrawal bleeding. As with other oral contraceptives, serious cardiovascular conditions such as venous thrombosis, pulmonary embolism, stroke, and myocardial infarction are rarely possible.
