Phenylpropanolamine

Products

Phenylpropanolamine is no longer on the market as a human drug in many countries. It was formerly contained in Astho-Med Syrup, Contac, Dimetane, Dimetapp, and Slim Caps, among others.

Structure and properties

Phenylpropanolamine (C9H13NO, Mr = 151.21 g/mol) has a phenylethylamine structure and exists as a racemate. The salt phenylpropanolamine hydrochloride is a white crystalline powder that is readily soluble in water. It is related to ephedrine and is also known as D,L-norephedrine.

Effects

Phenylpropanolamine (ATC R01BA01) has indirect sympathomimetic and appetite suppressant properties. Because of the possible adverse cardiovascular effects, the drug should not be taken in our opinion.

Indications

  • For short-term treatment of obesity during four weeks (no longer approved in many countries).
  • Rhinitis, common cold (no longer approved in many countries).
  • Urinary incontinence in dogs, as a veterinary medicine.

Abuse

Phenylpropanolamine is a centrally stimulating sympathomimetic and can be abused as an intoxicant or party drug, as well as a precursor chemical. Therefore, when dispensing veterinary drugs, it should be clarified whether the drug is actually intended for a dog.

Interactions

Drug-drug interactions are possible with guanethidine, other sympathomimetics, halothane, and MAO inhibitors.

Adverse effects

Possible adverse effects include rapid heartbeat, hypertension, cardiac arrhythmias, headache, irritability, drowsiness, nervousness, dizziness, urinary retention, and personality changes. Continuous use may lead to dependence. Phenylpropanolamine has been linked to serious cardiovascular disease such as stroke and heart attack.
