Rupatadine is commercially available in tablet and oral solution forms (Rupafin, Urtimed), among others. The drug has not yet been registered in many countries.
Structure and properties
Rupatadine (C26H26ClN3, Mr = 415.96 g/mol) is present in drugs as rupatadine fumarate. It is structurally related to loratadine. Rupatadine is also partially metabolized to desloratadine but is not a prodrug in the strict sense.
Effects
Rupatadine (ATC R06AX28) has antihistamine, mast cell-stabilizing, antiallergic, and PAF-antagonistic properties. The effects are due to dual histamine and PAF (platelet-activating factor) antagonism.
Indications
For symptomatic treatment of allergic diseases:
- Allergic rhinitis
- Urticaria
Dosage
According to the professional information. The tablets are usually taken once a day, regardless of meals.
Contraindications
Rupatadine is contraindicated in case of hypersensitivity. For complete precautions, see the drug label.
Interactions
Rupatadine is a substrate of CYP3A4 and corresponding drug-drug interactions are possible. Interactions with central depressant drugs and alcohol cannot be ruled out. A possible interaction with statins has been reported.
Adverse effects
The most common possible adverse effects include drowsiness, fatigue, headache, fatigue, dizziness, and dry mouth.
