Pyrvinium is commercially available as an oral suspension and in the form of dragées. It is no longer available in many countries.
Structure and properties
Pyrvinium (C26H28N3+, Mr = 382.5 g/mol) is present in pharmaceuticals as pyrvinium embonate or pyrvinium pamoate. Pyrvinium embonate is an orange-red to orange-brown powder with almost no odor and no taste that is practically insoluble in water. Pyrvinium embonate is photosensitive. It is a pyrrole and quinoline derivative and originally a dye.
Effects
Pyrvinium (ATC P02CX01) has antihelminthic activity against pinworms. The effects are due to inhibition of glucose uptake in the worms. Pyrvinium is poorly absorbed and acts locally in the digestive tract. Recent studies have shown that pyrvinium has additional antitumor properties.
Indications
Pinworm infestation. Unlike other anthelmintics, pyrvinium is approved only for this indication.
Dosage
According to the package insert. Pyrvinium is usually administered as a single dose independent of meals. A repeat dose may be given after 2-4 weeks if efficacy is insufficient.
Contraindications
Pyrvinium is contraindicated in hypersensitivity, liver impairment, inflammatory bowel disease, and renal insufficiency. For complete precautions, see the drug label.
Interactions
Interactions with other drugs have not been reported.
Adverse effects
Rarely to very rarely, allergic reactions, dizziness, cramps, and indigestion may occur. Because it is a dye, discoloration of, for example, stool, vomit, or clothing is possible.
