Afoxolaner is commercially available in the form of chewable tablets as a veterinary drug for dogs (NexGard). It was approved in many countries in 2015.
Structure and properties
Afoxolaner (C26H17ClF9N3O3, Mr = 625.9 g/mol) belongs to the isoxazoline group.
Effects
Afoxolaner (ATCvet QP53BX04) has insecticidal and acaricidal properties. Parasites ingest the active ingredient when they suck blood. The effects are due to inhibition of the GABA receptor (ligand-gated chloride channel). This results in neuronal hyperstimulation. Fleas are killed within 8 hours and ticks within 48 hours. The half-life is two weeks and the duration of action is approximately one month.
Indications
For the treatment of flea infestation, allergic flea dermatitis, and tick infestation in dogs.
Dosage
According to the product information leaflet. The dose is based on body weight. Chewable tablets are administered perorally once a month during the season. They may also be given with food.
Contraindications
Afoxolaner is contraindicated in cases of hypersensitivity. Refer to the drug label for complete precautions.
Adverse Effects
The most common potential adverse effect is vomiting.
