Efinaconazole is commercially available in the United States in the form of a solution (Jublia). The active ingredient has not yet been registered in many countries.
Structure and properties
Efinaconazole (C18H22F2N4O, Mr = 348.4 g/mol) structurally belongs to the triazole antifungals.
Effects
Efinaconazole has antifungal properties. The effects are due to inhibition of lanosterol 14α-demethylase. This inhibits the biosynthesis of ergosterol. Ergosterol is an important component of the fungal cell membrane. As with other agents, not all patients can be cleared of fungus with efinaconazole.
Indications
For local treatment of nail fungus of the toenails (caused by , ).
Dosage
According to the professional information. The solution is applied once daily for 48 weeks.
Contraindications
Efinaconazole is contraindicated in case of hypersensitivity. For complete precautions, see the drug label.
Interactions
Interactions with other drugs are not known. Efinaconazole is not a CYP450 inhibitor.
Adverse effects
The most common potential adverse effects include ingrown nails and local side effects such as rashes, vesicles, and pain.
