Nortriptyline

Nortriptyline was commercially available in the form of film-coated tablets (nortrilen). It was approved in many countries in 1964. It was discontinued from distribution in 2016.

Structure and properties

Nortriptyline (C19H21N, Mr = 263.4 g/mol) is present in drugs as nortriptyline hydrochloride, a white powder that is sparingly soluble in water. It is an active metabolite of amitriptyline (Saroten).

Effects

Nortriptyline (ATC N06AA10) has antidepressant, depressant, anticholinergic, central stimulant, and antihistamine properties. The effects are primarily due to inhibition of reuptake of norepinephrine into the presynaptic neuron. Nortriptyline is less sedating and anticholinergic than amitriptyline.

Indications

For the treatment of depressive disorders.

Dosage

According to the professional information leaflet. Tablets are usually administered one to three times daily. Treatment is started creeping in and stopped creeping out.

Adverse effects

The most common possible adverse effects include visual disturbances, headache, dizziness, tremor, dry mouth, constipation, nausea, fatigue, palpitations, and increased sweating.
