Pimozide is no longer commercially available as a finished drug in many countries. It is still available in some countries (Orap tablets).

Structure and properties

Pimozide (C28H29F2N3O, Mr = 461.5 g/mol) belongs to the diphenylbutylpiperidines. It exists as a white powder that is practically insoluble in water.

Effects

Pimozide (ATC N05AG02) is antipsychotic. The effects are probably due to the antidopaminergic properties.

Indications

For the treatment of chronic psychosis of the schizophrenic type.

Dosage

According to the professional information. Tablets may be taken once daily due to long half-life.

Adverse effects

Pimozide is metabolized mainly by CYP3A4 and may prolong the QT interval and rarely cause cardiac arrhythmias. This constellation is considered high-risk.
