Trientine was approved in many countries in 2020 in capsule form (Triogen).

Structure and properties

Trientine is present in the drug as trientine dihydrochloride, a white to slightly yellow hygroscopic powder that is readily soluble in water. It is triethylenetetramine.

Effects

Trientine (ATC A16AX12) forms a stable and soluble complex with copper. It promotes its excretion through the kidneys in the urine.

Indications

For the treatment of copper storage disease (Wilson’s disease) in patients who cannot tolerate treatment with D-penicillamine.

Dosage

According to the SmPC. Capsules are taken in 2 to 4 single doses 30 minutes to 1 hour before meals.

Interactions

Trientine lowers serum iron concentrations.

Adverse effects

Possible adverse effects include:

  • Nausea
  • Skin rash
  • Duodenitis, severe colitis
  • Anemia
  • Neurological deterioration
