Trifarotene was approved as a cream in the United States in 2019 and in Germany and many countries in 2020 (Aklief, Selgamis).
Structure and properties
Trifarotene (C29H33NO4, Mr = 459.6 g/mol) is a derivative of a terphenylic acid. It exists as a white to slightly yellow powder that is practically insoluble in water.
Effects
Trifarotene (ATC D10AD06) has comedolytic, anti-inflammatory, and antipigmentary properties. It affects cell differentiation and inflammation. The effects are due to selective agonism at the retinoic acid receptor (RAR). The binding affects gene expression. Trifarotene is selective for the γ-subtype of the receptor, which is found primarily in the skin. The absorbed drug is rapidly metabolized in the liver by CYP450 isozymes.
