Dapoxetine is commercially available in the form of film-coated tablets (Priligy). It has been approved in many countries since 2013.
Structure and properties
Dapoxetine (C21H23NO, Mr = 305.4 g/mol) is present in drugs as dapoxetine hydrochloride, a white powder with a bitter taste that is soluble in water. Dapoxetine is a naphthyloxyphenylpropanamine derivative. It has structural similarities to other SSRIs, such as fluoxetine.
Effects
Dapoxetine (ATC G04BX14) inhibits premature ejaculation. Effects are thought to be based on inhibition of neuronal serotonin reuptake.
Indications
For the treatment of premature ejaculation (ejaculatio praecox) in men 18 to 64 years of age.
Dosage
According to the drug label. Dapoxetine is not taken regularly, but as needed 1-3 hours before sexual activity. It is administered no more than once daily and may be swallowed with or without food.
Contraindications
Dapoxetine is contraindicated in hypersensitivity, certain cardiovascular diseases, mania, depression, and liver dysfunction. It should not be combined with MAO inhibitors, thioridazine, SSRIs, SNRIs, tricyclic antidepressants, and other agents with serotonergic activity, nor with CYP3A4 inhibitors. Interactions
Dapoxetine is predominantly biotransfected by CYP3A4, CYP2D6, and FMO1. Corresponding drug-drug interactions are possible. Dapoxetine has a high interaction potential (see also contraindications).
Adverse effects
The most common possible adverse effects include dizziness, headache, and nausea. Numerous other side effects are possible.
