Promestrien was commercially available in the form of vaginal capsules and as a vaginal cream (Colpotrophine). It was approved in many countries in 1982 and is now no longer available.
Structure and properties
Promestriene (C22H32O2, Mr = 328.5 g/mol) is an alkyl derivative of the natural estrogen estradiol.
Effects
Promestriene (ATC G03CA09) has estrogenic effects and promotes reconstruction of the atrophic vaginal epithelium. According to the SmPC, it is hardly absorbed into the organism and does not exert systemic effects.
Indications
For the treatment of vulvovaginal disorders secondary to estrogen deficiency.
Dosage
According to the SmPC. Capsules are administered locally once daily.
Contraindications
Promestrien is contraindicated in hypersensitivity, malignant changes of vulva, vagina and uterus, breast cancer, bloody discharge of unknown cause and during pregnancy. For complete precautions, see the drug label.
Interactions
No interactions with other drugs are known to date.
Adverse effects
Possible adverse effects include local burning and itching.
