Structure and properties
Anagrelide hydrochloride (C10H8Cl3N3O, Mr = 292.5 g/mol) is present in drugs as anagrelide hydrochloride monohydrate.
Effects
Anagrelide (ATC L01XX35) reduces platelet counts in peripheral blood. The mechanism of action is not precisely known but is probably related to megakaryocyte maturation.
Indications
For the treatment of essential thrombocythemia (increased number of platelets in the blood).
Dosage
According to the SmPC. Capsules should always be taken at the same time, either with a meal or fasting.
Contraindications
Anagrelide is contraindicated in hypersensitivity, moderate or severe liver impairment or renal insufficiency, and during pregnancy and lactation. For complete precautions, see the drug label.
Interactions
Drug-drug interactions have been described with phosphodiesterase inhibitors, antiplatelet agents, acetylsalicylic acid, and sucralfate.
Adverse effects
The most common possible adverse effects include diarrhea, abdominal pain, nausea, flatulence, headache, dizziness, palpitations, tachycardia, edema, and weakness.
