Structure and properties

Dasotralin (C16H15Cl2N, Mr = 292.2 g/mol) is a diastereomer of the desmethyl metabolite of sertraline (Zoloft, generics).

Effects

Dasotraline selectively inhibits the reuptake of the neurotransmitters dopamine and norepinephrine into presynaptic neurons in the central nervous system. The effects are due to inhibition of the corresponding transporters DAT and NET. Promotion of neurotransmission is associated with improvement of symptoms.

Indications

For the treatment of attention-deficit/hyperactivity disorder (ADHD). The agent is also being studied for the treatment of binge eating disorder.

Dosage

According to the SmPC. Because of its long half-life (47 to 77 hours), once-daily dosing is sufficient.

Abuse

Unlike amphetamines such as methylphenidate (e.g., Ritalin), this is not a stimulant and not a narcotic. The potential for abuse is considered low.

Adverse effects

The most common possible adverse effects include insomnia, poor appetite, and dry mouth.
