Lysine acetyl salicylate is commercially available as powder and injectable (Aspégic, Alcacyl powder, Germany: e.g., Aspirin i.v., Aspisol). It has been approved in many countries since 1973. Migpriv, which is combined with metoclopramide for migraine, was withdrawn from the market in many countries in December 2011 see under Migpriv. Structure and properties

Lysine acetyl salicylate (C15H22N2O6, Mr = 326.3 g/mol) is the salt of acetylsalicylic acid (aspirin) and the amino acid lysine. Lysine acetyl salicylate is a white crystalline powder that is more soluble in water than the sparingly soluble acetylsalicylic acid.

Effects

Like acetylsalicylic acid, lysine acetylsalicylate (ATC N02BA01) is analgesic, antipyretic, anti-inflammatory, and antiplatelet. The effects are due to inhibition of cyclooxygenase, which inhibits prostaglandin synthesis and the formation of thromboxane A2 inhibition. In contrast to acetylsalicylic acid, the onset of action is more rapid because of better water solubility, and lysine acetylsalicylate is also administered intravenously. The maximum plasma concentration is reached within 15 minutes.

Indications

For the treatment of pain and fever of various causes. As an antiplatelet agent (Kardégic 100, 300).

Dosage

According to the drug label. The powder should be taken with sufficient liquid and, if possible, after a meal.

Contraindications

  • Hypersensitivity, including to acetylsalicylic acid, salicylates, or NSAIDs.
  • Gastric or duodenal ulcer
  • Coagulation disorders
  • Severe disorders of liver or kidney function
  • Last 3 months of pregnancy

Adverse effects

The most common possible adverse effects include microbleeding, stomach cramps, dyspepsia, nausea, and vomiting. Rarely, serious adverse reactions such as gastric bleeding, asthma, hypersensitivity reactions, and renal dysfunction are observed. Lysine acetyl salicylate prolongs bleeding time.
